Uracil-DNA
Glycosylase is an enzyme that catalyzes the hydrolysis of the N-glycosyl bond
between uracil and the sugar residue in single- and double-stranded DNA. The
enzyme possesses maximal activity at 37°C. Uracil-DNA Glycosylase is used in
the preparation of PCR mixtures to prevent the appearance of false-positive
results caused by contamination of the PCR reaction mixture with products of
previously performed PCR (amplicons), which are destroyed before amplification.
The enzyme is not active against RNA and short oligonucleotides. The enzyme is
inactivated completely during the first cycle of DNA denaturation and does not
interfere with the current reaction amplification products.
